It pretty simple, really.  The most common method that's used is the Jacob's Method.  They take a picture of the entire area where there are people gathered, then make a grid over them.  They count the number of people in one of the grids, then multiply that number by the number of grids over the crowd.  I remember that explanation from the Million Man March in D.C. back in the mid-90's.  Here's a more precise explanation:

If anything I'm always rooting for kate but it's amazing how old she looked in the white dress today , I mean when I'm that age I want to look just as good and she's very pretty but it was sooo ageing the way she did her hair and make up idk this entire tour has been kate dressing like a school teacher attending a prize day ..sorry but it's just what I noticed .

The Duke of Cambridge thanks Australia for the "wonderful memories" and reveals Prince George's personal tour highlight.
